Porcine Hoof And Human Toenail Cultures
Porcine hooves obtained from Yorkshire pigs were used as an ex vivo model for human nail. The protocol to use Yorkshire pigs was approved by the Subcommittee on Research Animal Care at Massachusetts General Hospital. The hooves were sliced into square fragments of approximately 0.7 × 0.7 cm. Human toenail clippings were also collected from anonymous volunteers without onychomycosis. The toenail clippings were cut into circular fragments with 4-mm diameter tissue biopsy punches. The thickness of the hoof and toenail fragments ranged from 0·80 to 1·66 mm and from 0·51 to 1·18 mm, respectively, determined by a digital Vernier caliper.
Prior to being autoclaved at 120 °C for 15 min, the hoof or toenail fragments were sterilized by immersion in 70% isopropanol twice for 1 min, and then washed twice with sterile water. Initially fragments were inoculated on the ventral surface with 20 L T. rubrum suspension containing approximately 2·0 × 10 or 2·0 × 10 colony-forming units mL1, and later they were immersed in 100 L T. rubrum suspension containing 2·0 × 106 c.f.u. mL1 or 2 h. The fragments were then placed in 35-mm diameter Petri dishes, which were placed inside a 150-mm diameter Petri dish containing sterile water to provide a moist environment. The dishes with hoof or toenail fragments were then incubated at 30 °C for 28 weeks before the experiments were performed.

Once youve purchased a UV-C lamp, your next step is to identify all of the surfaces that are currently infested with the mold fungus. Any surfaces you miss could allow the mold to live on and continue spreading throughout your home or business.
Once youve determined which surfaces have mold on them, you should keep the UV-C lamp around two inches away from the mold in question. Every other light source in the room will need to be turned off in order for the UV light to get rid of all of the mold. This includes any doors and windows that could let small amounts of light in. The mice were imaged with the luminescence camera immediately after the application of the C. albicans to ensure that the fungal inoculum applied to each burn was consistent.
